TGFBR2 gene silencing inhibits proliferation and induces apoptosis of osteoblasts

AIM: The aim of this paper was to explore the role of transforming growth factorβ (TGF-β) in regulating osteoblasts via the type I and II receptors.
METHODS: In the current study, we constructed an lentivirus shRNA targeting TGFBR2 to inhibit the expression of TGF-β, and demonstrated its effect on the proliferation, apoptosis, and functions of osteoblasts, as well as the mechanism involved.
RESULTS: Our data indicated that the growth of TGFBR2 knockdown osteoblasts was inhibited significantly. Moreover, the ALP staining of osteoblasts showed that the function of the osteoblast with TGFBR2 gene silencing was also affected in vitro. In addition, FCW detection showed that the apoptosis of osteoblasts increased, and the expression of p-Smad2 also increased.
CONCLUSION: All the results indicated that TGFBR2 genes played a significance role in regulating the growth of osteoblasts and maintaining its function via the Smad2 signaling pathway. The results shed the light on understanding the bone growth process and pathology of related disease such as osteoporosis.
