Isolation of endophytic fungus producing baccatin III from Taxus wallichiana var. mairei

AIM: The objective of this study was to isolate endophytic fungi producing baccatin III from yew for the sake of baccatin III and taxol manufacture.
METHODS: The surface sterilized bark of Taxus wallichiana var. mairei. was taken as material and PDA culture medium was used in the experiment. The fungal culture was extracted with mixture of chloroform / methanol (1:1, v/v) and the baccatin III in extract was determined and authenticated with LC-MS. The endophytic fungus was identified with ITS rDNA and 26S D1/D2 rDNA sequence.
RESULTS: The results showed that total of 192 endophytic fungus strains were isolated from T. wallichiana var. mairei. The strain producing baccatin III was screened from 192 strains and identified as Diaporthe phaseolorum. The productivity of this strain cultured in PDA culture medium amounts to 0.219 mg/L.
CONCLUSION: The isolated endophytic fungus is high in yield and prospective to be applied in baccatin III and taxol manufacture after improving the condition of culture or breeding.
