A collection of expressed sequence tags (ESTs) from totipotent cDNA of durum wheat

Aim. This study aims to the development of a database to store our expressed sequence tags (ESTs). The sequencing of a cDNA library to generate ESTs is an effective tool for research activities such as gene discovery, generation of molecular markers and characterization of transcription patterns. In this issue we report a sequence analysis of a durum wheat cDNA library.
Methods. In order to maximize the mRNAs representativeness, a patented method is used, which allows the preparation of a totipotent cDNA library from durum wheat regardless of the developmental stage of the material from which the library is originated.
Results. To date, we have archived 11 000 ESTs and 7 648 of them were subjected to both a BLASTX and BLASTN search.
Conclusion. When compared to public databases, 94%, 1% and 5% of the ESTs sequences showed similarity, respectively, to genes from Triticum aestivum, Triticum turgidum and other Triticeae, approximately other organisms and no match to known sequences. Moreover, we created a functional inventory of these sequences based on the BLASTX annotation.
