Correlation between mRNA tissue expression and serum concentration of Cyr61 in adenomyosis

BACKGROUND: Adenomyosis is a disease characterized by heterotrophic stromal and endometrial glands within myometrium surrounded by myometrium hyperplasia. Alterations of neo-angiogenesis system contributed to adenomyosis pathogenesis. One of the protein roles in adenomyosis pathophysiology is Cysteine-rich 61 (Cyr61). However, its relation to adenomyosis has not been determined as far. The study was aimed to investigate the expression of Cyr61 in adenomyosis and without adenomyosis and determine the correlation between mRNA tissue expression and serum concentration of Cyr61 in patients with adenomyosis.
METHODS: The research was conducted at Hasanuddin University hospital and the network hospitals using a cross-sectional study. The samples were 30 women including 15 women with histologically diagnosed adenomyosis and 15 women with histologically diagnosed without adenomyosis and endometriosis, who underwent a hysterectomy. Samples of serum and endometrial tissue were collected, and Cyr61 mRNA levels were determined by RT-PCR. The Cyr61 protein levels in endometrial tissue were determined by immunohistochemistry by ELISA. The data were analyzed using the Chi-square test, Mann Whitney test, and Spearman Correlation test with the significance P=0.05.
RESULTS: The expression Cyr61 was higher in the patients with adenomyosis than in the patient without adenomyosis like myoma uterine or cyct ovarium. Cyr61 expression in the endometrial tissue has a positive correlation with the serum concentration (P=0.001).
CONCLUSIONS: The expression of Cyr61 was involved in the pathogenesis of adenomyosis and there is a correlation between expression mRNA Cyr61 in endometrial tissue and serum concentration in adenomyosis patients.
