Association between -308(G/A) and -1031(T/C) polymorphism in tumor necrosis factor alpha among polycystic ovarian syndrome patients

BACKGROUND: TNF-α is a pro-inflammatory cytokine which has many immunological relations with many gynecological diseases. -308(G/A) and -1031 T/C polymorphism in the promoter region of the TNF-α gene is reported in many gynecological diseases and from these polycystic ovarian syndrome (PCOS). The aim of this study is to study the association between -308(G/A) and -1031 T/C polymorphism in the promoter region of the TNF-α gene among PCOS Jordanian females.
METHODS: -308(G/A) and -1031 T/C polymorphism in the promoter region of the TNF-α gene is determined by restriction fragment length polymorphism (RFLP). One hundred and fifty PCOS patients and on hundred and sixty age matched control were enrolled in this study. DNA was extracted using the Wizard Genomic DNA Purification kit, amplified TNF-α gene by PCR and restriction digested to determine -308(G/A) and -1031 T/C TNF-α genotype. Statistical analysis was performed using the Statistical Package for Social Sciences (SPSS).
RESULTS: The results of this study showed an association between CC genotype of -1031(T/C) of TNF-α and PCOS among Jordanian females. This study demonstrated a strong significant difference between -1031C alleles among PCOS and control patients (P=0.026), and that the C allele was significantly higher among PCOS patients.
CONCLUSIONS: -1031 T/C polymorphism in the promoter region of the TNF-α gene was associated with PCOS which may be considered as a marker for PCOS diagnosis.
