A comparative study of simultaneous determination by capillary zone electrophoresis and high performance liquid chromatography of allantoin, uric acid, hypoxanthine and xanthine in atherosclerotic plaque

Background. Allantoin, uric acid (UA), hypoxanthine (Hx) and xanthine (X) are enhanced during oxidative stress. We want determine these metabolites in atherosclerotic plaque, in which oxidative stress is known, to improve our knowledge on atherosclerosis and to give a further help in the plaque characterization. In this preliminary work, we propose a comparative study for the simultaneous determination of these compounds by capillary zone electrophoresis (CE) and high performance liquid chromatography (HPLC), to evaluate the method most suitable for this analysis. This study is made both using standards and carotid plaque.
Methods. Surgical specimens were obtained from patients undergoing carotid endarterectomy. Electrophoretic separations were performed a Waters Quanta 4000 instrument; for HPLC separation a Varian Vista 5500.
Results. The HPLC method enables resolution in gradient of the four compounds in 30 min, while CE method in 20 min.
Conclusions. The CE and HPLC separations of allantoin, UA, Hx and X may be used for a satisfactory determination in vivo, in short time. Moreover the CE separation gave more precise allantoin dosage, even when the concentration is low.
