Study of the impact of Id-1 on human colon carcinoma cell line SW480 proliferation and migration

BACKGROUND: This study aims to explore the effect of inhibitor of DNA binding-1 (Id-1) on human colon carcinoma cell line SW480 proliferation and migration.
METHODS: SW480 cells transfected with Id-1-interferrence sequence were assigned as the inhibition group, and SW480 cells with blank interference sequence (blank group) and blank vector transfection (empty vector group) were assigned as the control group. Id-1 mRNA and protein expression in these three groups were detected by reverse transcription-polymerase chain reaction (RT-PCR) and western blot. Cell proliferation in these three groups was assessed by MTT assay. Effect of the proliferation and migration of Id-1 on SW480 cells was observed using the wound healing assay.
RESULTS: RT-PCR and western blot results revealed that the mRNA and protein expression of Id-1 in the inhibition group was lower than that in the blank group and empty vector group (all, P<0.01). The results of the growth curve revealed that proliferation ability was significantly weaker in the inhibition group than in the blank group and empty vector group from the third day. The wound healing assay revealed that the migration velocity was slower in the inhibition group than in the blank group and empty vector group. However, there was no difference between the blank group and empty vector group. The Matrigel invasion experiment revealed that the number of Transwell cells decreased significantly in the inhibition group than in the blank group and empty vector group (P<0.01).
CONCLUSIONS: Id-1 plays an important role in the proliferation and migration of human colon carcinoma cell line SW480.
