Molecular epidemiology of ompP5 in Haemophilus influenzae spp in clinical isolates

AIM: Protein 5 (P5) is one of the Haemophilus influenzae OMPs that expressed by H. influenzae type b (Hib) and Nontypeable H. influenzae (NTHi). The purpose of this research was to determine of molecular epidemiology of the ompP5 in H. influenzae spp in clinical isolates of H. influenzae.
METHODS: This study involved 21 Haemophilus influenzae strains isolated from patients in two Tehran hospitals. Restriction enzyme analysis was performed on the 15 clinical samples using HinfI and AluI. By detecting the subsequence and comparing the subsequences of ompP5 gene of 5 clinical strains of H. influenzae with standard strain, the similarities and differences between them were found.
RESULTS: Conserved and variable region of ompP5 subsequently was amplified in 17 and 16 samples from all 21 clinical samples. Hinfl restriction pattern in 14 samples out of 15 were similar and in 1 sample was different; Alul restriction pattern in 11 samples out of 15 were similar and in 4 samples were different from the standard strain (Hib). The results of subsequence detection for the ompP5 conserved region of samples 1,2,4,5 and 6 had 96.88%, 100%, 99.22%, 99.02% 96.88% similarity with conserved region of Hib ompP5 sequence; this similarity for this samples variable region was 86.24%, 100%, 92.56%, 93.94% and 88.24%.
CONCLUSION: ompP5 gene was observed in 18 sample out of 21 clinical samples and had different in sequences of both conserved and variable regions in respect to each other and to the H. influenzae strains that be submitted in NCBI.
