Circulation monocyte chemoattractant protein-1 levels on Wistar Rat induced obesity with high fat diet

BACKGROUND: High fat diet (HFD) causes inflammation and obesity. Obesity was characterized as low-grade systemic inflammation. The purpose of this research was to identify the effects of obesity-induced high-fat-diet on systemic inflammation with biomarkers of circulation monocyte chemoattractant protein-1 (MCP-1) levels.
METHODS: 30 male wistar rats aged 10-12 weeks (body weight between 170-220 grams) were randomly divided into 5 groups (6 rats/group): baseline, normal diet for 8 weeks (ND8), ND for 16 weeks (ND16), HFD for 8 weeks (HFD8) and HFD for 16 weeks (HFD16). Body weight and naso-anal length were measured to get the index value of obesity and body fat percentage. Obesity index measured were Lee Index, Rohrer Index and TM Index. Blood samples obtained by intracardiac for examination of Circulation MCP-1 levels using Elisa method.
RESULTS: The administration of HFD for 8 and 16 weeks increased body weight, obesity index, body fat percentage and circulation MCP- levels. Circulation MCP-1 levels in HFD group was higher than ND group both between ND8 and HFD8 and between ND16 and HFD16. The longer provision of HFD, the higher circulation MCP-1 level. Body weight, obesity index, and body fat percentage are positively correlated with circulation MCP-1 levels.
CONCLUSIONS: Obesity-induced high-fat diets caused systemic inflammation that was characterized by increased levels of MCP-1 circulation.
