The effect of histone deacetylase inhibitor on the restoration of atrophying aged skeletal muscles

BACKGROUND: The purpose of this study is to investigate the effects of trichostatin A (TSA) on disorganized sarcomeric integrity/remodeling in denervation-induced atrophy of aged skeletal muscles.
METHODS: The effect of TSA on denervated muscles was investigated: YNcon, young normal control; ANcon, aged normal control; YDcon, young denervated control; ADcon, aged denervated control; YDinj, young denervated TSA injection; ADinj, aged denervated TSA injection. The bilateral hindlimb muscles were obtained 4 weeks after injury.
RESULTS: The ADinj had an increased pattern of MWW in relation to ADcon in GCM muscle. Morphological analysis showed smaller round fibers in YDcon and ADcon. However, degraded muscle fibers in ADcon (35% decrease in relation to ANcon) and ADinj (45% in relation to ANcon) was more remarkably decreased than in each corresponding young group.
CONCLUSIONS: Despite aging and muscle types-dependent effect in ADcon and YDcon, we suggest that denervation remarkably affects in both aged and young group. We could not find the preventive TSA effect from denervation-derived muscle atrophy, however, TSA effect at micro level such as fiber size or fiber type shift can be carefully anticipated. This suggests that it needs to find the way like physical activity for preventing muscle atrophy in case of recovery from denervation within a short period.
