Hearing loss is a common sequela to many inner ear disorders. However, the therapeutic options for these conditions are limited as the inner ear is inaccessible to many drugs. Theoretically, nanoparticles that permeate the round window can target specific cell populations and provide controlled drug release. Nanoparticles have shown promise as an inner ear drug delivery vehicle in many reports. Toxicity, especially with long-term use, should be determined prior to clinical use. Also, the drug to nanoparticle loading ratio should be increased to maximize the delivery efficacy and minimize toxic effects. Understanding of the RW, including the mechanism of passage, should be expanded.
