AIM: The aim of this study was to observe RhD-negative patients for the RHD gene polymorphism in the Han population of Xuzhou, China. The RhD negative trait is generated by multiple genetic mechanisms and has been shown to be dependent based on the ethnic group.
METHODS: In this study, conventional serological detection methods and the antiglobulin test (IAT) were used to confirm the RhD antigen phenotype. We used the sequence specific primers polymerase chain reaction (PCR SSP) method to detect the RHD gene.
RESULTS: There were 110 RhD negative individuals. The PCR detection of RhD gene results showed that there were five cases of RhD positives, 47 cases of RH negatives, 22 cases of RH-CE(2-9)-D genetic, 13 cases of the DVI III genes, and two cases had the weak D15 gene, and 17 cases had the DEL RHD1227A gene.
CONCLUSION: The complex structure of RhD negative individual with the RhD gene polymorphisms is present in the Han population of Xuzhou, China. The variant alleles RH-CE (2-9)-D gene, DVI III, DEL-1227A gene were the most dominant.
