Genechip technology for the study of complex systems. The example of dendritic cells

The appli­ca­tion of ­high den­sity DNA ­array tech­nol­o­gy to mon­i­tor ­gene expres­sion tran­scrip­tion has ­been respon­sible of a par­a­digm ­shift in biol­o­gy and we ­have now the abil­ity to meas­ure the expres­sion of a sig­nif­i­cant pro­por­tion of the ­genome in sin­gle experi­ment. In the ­field of immu­nol­o­gy the anal­y­sis of ­gene expres­sion pro­files has ­revealed ­that ­cells ­respond to an acti­va­tion sig­nal ­with ­waves of coor­di­nat­ed ­gene expres­sion pro­files. DCs rep­re­sent the pro­fes­sion­al anti­gen pre­sent­ing ­cells ­which ­uptake, pro­cess and ­present anti­gens to ­naive T ­cells induc­ing ­their prim­ing. In ­order to ­acquire ­this abil­ity DC ­have to under­go a pro­cess of mat­u­ra­tion ­that ­involve the acti­va­tion of hun­dreds of ­genes. ­With the ­attempt to inves­ti­gate dif­fer­enc­es in DC mat­u­ra­tion ­induced by dis­tinct dif­fe­ren­ti­a­tion stim­u­li, we com­pared the tran­scrip­tion­al pro­gram of ­mature and imma­ture DC, ­after ­either LPS or ­TNFα stim­u­la­tion ­using a Gene­Chip oli­go­nu­cleo­tide micro­ar­ray ­which per­mits the simul­ta­ne­ous anal­y­sis of the expres­sion of 6500 ­genes and ­ESTs. A ­very ­diverse mod­ula­tion of ­gene expres­sion was ­observed in the two cas­es. ­Only LPS-stim­u­lat­ed ­cells ­showed a pat­tern of expres­sion of ­genes com­pat­ible ­with a defin­i­tive ­growth ­arrest and ­with a suit­able acti­va­tion and con­trol of the ­immune ­response.
