Genechip technology for the study of complex systems. The example of dendritic cells

The application of high density DNA array technology to monitor gene expression transcription has been responsible of a paradigm shift in biology and we have now the ability to measure the expression of a significant proportion of the genome in single experiment. In the field of immunology the analysis of gene expression profiles has revealed that cells respond to an activation signal with waves of coordinated gene expression profiles. DCs represent the professional antigen presenting cells which uptake, process and present antigens to naive T cells inducing their priming. In order to acquire this ability DC have to undergo a process of maturation that involve the activation of hundreds of genes. With the attempt to investigate differences in DC maturation induced by distinct differentiation stimuli, we compared the transcriptional program of mature and immature DC, after either LPS or TNFα stimulation using a GeneChip oligonucleotide microarray which permits the simultaneous analysis of the expression of 6500 genes and ESTs. A very diverse modulation of gene expression was observed in the two cases. Only LPS-stimulated cells showed a pattern of expression of genes compatible with a definitive growth arrest and with a suitable activation and control of the immune response.
