Aim and methods. A total of ten personal tools, rapresented by five wallets and five towels, was typed on blind to evaluate the success rate in DNA recovery. An appropriate sampling method was used to collect the biological material from the objects investigated: surfaces of the wallets were swabbed using sterile swab, wet with sterile water; a small fragment of the fabric (approssimatively 20 mm2) was sampled to collect the material from the towels. The items analyzed were used by five subjects; an oral swab from every subject tested was collected in order to compare the genetic profiles obtained from the material analyzed. Samples collected from the items were amplified using NGM™ PCR Amplification Kit using the analytical conditions reported in the user’s manual. Also, we evaluated the Lewis typing from a blood sample collected from everyone subject tested to understand if it was involved in an increased body secretions.
Results. We have shown that there are differences between individuals in their tendency to deposit DNA on an item when it is touched.
Conclusion. The findings reported here are preliminary and further investigations are underway in order to further add to understanding of the issues of DNA transfer and persistence.
