AIM AND METHODS: Medium components were optimized using one-factor-at-a-time and orthogonal array method in this study. With biological potency as index, the best carbon source and nitrogen source respectively were soluble starch and yeast extracted through one-factor-at-a-time test. Subsequently, the concentration of soluble starch, yeast extract and mineral sources were optimized using the orthogonal array method.
RESULTS AND CONCLUSION: The results showed that the optimal culture medium was 7.0% soluble starch, 2.5% yeast extract, 0.6% NaCl, 0.2% MgSO4 and 0.2% K2HPO4, and the order of factor effects on biological potency of supernatant of strain YS-1 was soluble starch >MgSO4 > yeast extract >K2HPO4 >NaCl. In addition, the confirmation experiments determined the validity of the method. Biological potency of supernatant of strain YS-1 in optimized media by orthogonal array methods was about 2.53 times greater than higher than that of one-factor-at-a-time experiment, and 4.52 times greater than with the basal medium.
